Key Factors Driving the Growth of the Lupus Nephritis Market

  • Increasing Prevalence of Systemic Lupus Erythematosus: The growing prevalence of systemic lupus erythematosus (SLE), a significant proportion of whom develop lupus nephritis, is expanding the patient population requiring treatment. Improved disease recognition and diagnosis are also contributing to greater identification of eligible patients.
  • Growing Demand for Targeted and Effective Therapies: The limitations and safety concerns associated with conventional immunosuppressive treatments have created a need for more targeted therapies. Novel biologics and other advanced treatment approaches are helping address unmet needs in patients with active or difficult-to-manage disease.
  • Advancements in the Clinical Pipeline: A robust pipeline of investigational therapies, including Anifrolumab (SAPHNELO; AstraZeneca), Ianalumab (VAY736; Novartis/MorphoSys), Cenerimod (Idorsia Pharmaceuticals/Viatris), Rapcabtagene autoleucel (YTB323; Novartis), Iptacopan (FABHALTA; Novartis), and others with diverse mechanisms of action, is expected to reshape the lupus nephritis treatment landscape. Ongoing clinical trials and positive study outcomes could introduce new treatment options and intensify market competition.
  • Growing Interest in CAR-T Therapy for Lupus Nephritis: The success of CAR-T therapies in oncology has accelerated interest in their application to autoimmune diseases, including lupus nephritis. Early clinical evidence demonstrating deep B-cell depletion and potential long-term disease control has attracted substantial investment, positioning CAR-T therapy as a promising future disease-modifying approach.
  • Rising Focus on Personalized Treatment Approaches: The heterogeneity of lupus nephritis is driving interest in personalized and targeted treatment strategies. Advances in biomarkers and disease monitoring may help optimize therapy selection and improve long-term patient outcomes.

Aparna Thakur, project manager of forecasting at DelveInsight, said that all CAR-T therapies, C-CAR168, resecabtagene autoleucel, and rapcabtagene autoleucel, obecabtagene autoleucel show promise for treating lupus nephritis, but it is too early to definitively say which is better.

Lupus Nephritis Market Analysis

  • At present, three therapies have received FDA approval for the treatment of lupus nephritis: belimumab (BENLYSTA), approved in 2020 and subsequently extended to pediatric patients in 2022; voclosporin (LUPKYNIS), approved in 2021 as the first oral therapy specifically indicated for lupus nephritis; and obinutuzumab (GAZYVA/GAZYVARO), approved in 2025, broadening the range of biologic treatment options available for patients with active disease.
  • Despite these advances, the existing standard of care for lupus nephritis continues to rely largely on broad-spectrum immunosuppressive agents, which are associated with considerable short- and long-term toxicity.
  • Growing research efforts have focused on developing targeted therapies designed to modulate specific inflammatory and immune pathways implicated in disease progression.
  • The lupus nephritis market has also attracted increasing interest from major pharmaceutical companies.
  • The evolving drug pipeline includes several innovative therapeutic approaches, such as complement inhibitors like iptacopan, BAFF-R antagonists such as ianalumab, monoclonal antibodies including anifrolumab, sphingosine-1-phosphate 1 receptor modulators such as cenerimod, and CAR-T therapies including resecabtagene autoleucel, rapcabtagene autoleucel, obecabtagene autoleucel, C-CAR168, and mivocabtagene autoleucel.
  • These emerging therapies are anticipated to contribute to significant changes in the lupus nephritis treatment landscape through 2036, potentially enabling more targeted, personalized, and effective disease management.

Lupus Nephritis Competitive Landscape

Some of the lupus nephritis drugs under development include Anifrolumab (SAPHNELO; AstraZeneca), Ianalumab (VAY736; Novartis/MorphoSys), Cenerimod (Idorsia Pharmaceuticals/Viatris), Rapcabtagene autoleucel (YTB323; Novartis), Iptacopan (FABHALTA; Novartis), Obecabtagene autoleucel (obe-cel; Autolus), C-CAR168 (AbelZeta), Resecabtagene autoleucel (CABA-201; Cabaletta Bio), FT819 (Fate Therapeutics), and others.

AstraZeneca’s SAPHNELO (anifrolumab) is an intravenous, fully human monoclonal antibody that targets Subunit 1 of the Type I interferon (IFN) receptor. By binding to this receptor, the therapy inhibits Type I IFN signaling. Type I IFNs, including IFN-alpha, IFN-beta, and IFN-kappa, are cytokines that play an important role in the regulation of inflammatory responses.

AstraZeneca is currently assessing SAPHNELO in the Phase III IRIS trial (NCT05138133) for adults with active proliferative lupus nephritis. According to the company’s 2024 full-year results update, topline results from the Phase III IRIS study are expected in 2026.

Fate Therapeutics’ FT819 is an allogeneic, off-the-shelf CD19-directed CAR T-cell therapy candidate designed to enhance both safety and therapeutic efficacy. Fate Therapeutics has initiated the Phase II RECLAIM-LN study and treated its first patient to evaluate FT819 in individuals with moderate-to-severe systemic lupus erythematosus (SLE) accompanied by Class III or IV lupus nephritis.

The open-label trial is expected to enroll approximately 53 patients and will evaluate the safety and efficacy of a single 900-million-cell dose of FT819. The primary endpoint is the percentage of patients achieving a complete renal response at Week 26. The study incorporates recommendations provided by the US FDA as part of FT819’s Regenerative Medicine Advanced Therapy (RMAT) designation.

Novartis’ Rapcabtagene autoleucel is an investigational autologous CD19-directed CAR T-cell therapy developed on Novartis’s T-Charge platform. The treatment uses a patient’s own T cells, which are genetically modified to express a CD19-specific chimeric antigen receptor (CAR), allowing selective targeting and depletion of B cells involved in autoimmune disease. Delivered as a single intravenous infusion, rapcabtagene autoleucel is intended to achieve profound and durable B-cell depletion.

Novartis is currently investigating rapcabtagene autoleucel in a Phase II clinical study for SLE and lupus nephritis. Based on Novartis’s 2026 interim financial report, the company expects to submit regulatory applications for rapcabtagene autoleucel in lupus nephritis/SLE in 2028.

The anticipated launch of these emerging lupus nephritis therapies are poised to transform the lupus nephritis market landscape in the coming years. As these cutting-edge lupus nephritis therapies continue to mature and gain regulatory approval, they are expected to reshape the lupus nephritis market landscape, offering new standards of care and unlocking opportunities for medical innovation and economic growth.
